Allergen-PTs (qualitative / quantitative with z-scores and recovery rates) - Accredited Programs -

The PROCESS of PT rounds ptAL01 - 09 + 13: 

Two Samples (25g each) with the Allergens in a Food Matrix (Samples A + B), as cheese, pastry, cocoa spread or boiled sausage are provided for qualitative and quantitative determination. One of these samples contains the spiked or incurred allergen(s). Incurred allergens were added at the same time as the other food matrix ingredients and processed in the matrix (e.g. baked or cooked).
The
third sample, the "Spiking Level Sample" (Sample SP) (15g) contains the same allergens in potato powder, a simple matrix without interfering compounds and without further processing in similar amounts (at the „spiking level“). 

The evaluation of results will be qualitative and quantitative. Quantitative statistics with z-scores will be done with the target values from the consensus values of partcipants' results (5 results) and from the known gravimetrically spiked values.
Recovery rates and recovery z-scores are calculated for every single quantitative result of the "positive"-samples and the "Spiking Level Samples", regardless which method was applied. By comparing the recovery rates in tabular form, the influence of the food matrix and processing on the performance of the analytical method can be directly seen.

Any suitable method can be applied for analysis, in particular ELISA, LFD, PCR and LC-MS.

Allergen-Screening (qualitative and semi-quantitative) 

ptALS1-3: 4 Samples (20g each) with Allergens in a simple Food Matrix (rice flour) are provided. All parameters are contained in 2 of 4 samples providing 2/3 and 1/6 of the new Action Levels (VITAL 4.0, 2024), respectively. The “action level” in mg/kg is calculated from the allergen protein reference dose per 100 g food portion. The positive detection of the 2/3 action level ensures that the action level can be detected safely even when the measurement uncertainty of the applied method is about 50%. While the 1/6 action level can proof the method performance at lower levels close to the LOQ.
The evaluation of results is strictly qualitative with indication of the obtained agreements with the consensus values of the participants and the spiking of samples. All gravimetrically spiked quantitative levels are given in the reports which enables a semi-quantitative estimation of results..
For analysis ELISA (+ Lateral Flow), PCR and LC/MS can be applied.

Allergen Swab Test (qualitative) - Wipe Test for Surfaces

ptASW1-3: There are 2 samples of a Test Surface with Allergens offered for the detection of each allergen by means of a wipe test (swab test). The allergens to be tested were attached to one test surface, while the other test surface is "negative". The evaluation is exclusively qualitative (positive/negative) with indication of the obtained agreement with the consensus values of the participants and the spiking of the surfaces. As an analytical test method, the Lateral-Flow-Test is particularly suitable. ELISA, PCR and LC/MS can also be used. Multiple sets of results can be submitted for different methods.
Please note: When different (extraction) methods are applied, further sample sets are required. Double sample material can be ordered.

Single Proficiency Test (SPT) Program of all DLA Allergen Response PTs and Action Level PTs
These SPTs are
especially suitable for method validation and verification!

By means of a Single Proficiency Test it is possible to participate individually and flexibly at any time, even outside the regular PT program, in a proficiency test format for a single participant laboratory. The target values (assigned values) of the SPT test material are determined on the basis of known reference values from previous PTs / inter laboratory tests, contents of spiked analytes (reference standard) or by reference laboratories.





Allergen-Response SPT of processed allergenic Ingredients

Action Level Matrix- ALM Verification

sptALM1-11: A kind of Calibration Set of 5 Samples (20g each) of an Allergen in a Specific Matrix is provided for qualitative determination as well as a „Blank Sample“ (Matrix). The allergen levels of the calibration row range from 1/10 to at least twice the Action Level 1 (VITAL concept 3.0) and judgment value of the German Commission ALTS/ALS. Valuation of PT-results is qualitative in Scores of 1-5 (Score 3 = Action Level successfully identified). Quantitative results will be reported for information. The analytical methods are optional.

4. Genitically modified Organisms






GMO-Screening I and II (qualitative) 

ptGMS1 + ptGMS2: 4 samples (10g each) with GMO-screening sequences / “markers” (e.g. promoter- and terminator sequences, herbicide or insect resistance genes) and GMO events in different matrices (flour and semolina mixtures) are provided.
All mentioned parameters are contained in at least one of the four samples.
The evaluation of results is strictly qualitative with indication of the obtained agreements with the consensus values of the participants and the spiking of samples. All gravimetrically spiked quantitative levels are given in the reports for further information.
For analysis PCR, Real Time PCR, ELISA (+ Lateral Flow) can be applied.
Multiple sets of results can be submitted for different methods.
